David Miller hit an undefeated 68 off 38 balls, including three sixes in the first three balls of the match last over.
Miller and skipper Hardik Pandya shared an unbroken 106-run fourth-wicket partnership in only 61 balls.
Pandya finished the game undefeated with 40 from 27 balls.
Earlier, Jos Buttler's 89 off 56 balls and Sanju Samson's 47 (26) boosted Rajasthan Royals to a first-innings lead of 188/6.
In the second qualifier, RR will meet the winner of the eliminator between Royal Challengers Bangalore and Lucknow Super Giants.
On Wednesday, eliminator will be performed at Eden Gardens.
The second qualifier will be held on May 27 at the Narendra Modi Stadium in Ahmedabad.
